Incubation<\/strong><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Place the tube at a 45-degree angle and incubate it for 1 hour at 38.5\u00b0C. During this time, the most active sperm will swim upwards into the medium.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>5. Collection of the Supernatant<\/strong><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">After incubation, gently extract approximately 750 \u00b5l of the supernatant. This layer contains the most mobile, high-quality sperm.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>6. Sperm Concentration<\/strong><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">&#8211; Transfer the supernatant to a clean tube and centrifuge it at 300 rcf for 7 minutes.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">&#8211; Discard 650 \u00b5l of the liquid, leaving 100 \u00b5l of the pellet. This pellet contains the selected sperm.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>7. Final Adjustments<\/strong><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">&#8211; Homogenize the pellet gently and take 10 \u00b5l to mix with 990 \u00b5l of formol saline. This mixture allows for accurate sperm counting using a Neubauer chamber.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">&#8211; Adjust the final concentration to 1&#215;10\u2076 sperm\/ml to prepare for insemination.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>8. Insemination<\/strong><\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Use the prepared sample to inseminate each well, ensuring a consistent final concentration.
